Glucophage 500mg Tablet (Pack of 50) is an oral antidiabetic medicine by Merck containing metformin hydrochloride 500mg per film-coated tablet, prescribed as a first-line treatment for type 2 diabetes mellitus (T2DM) in adults and children aged 10 years and above. How It Works

Metformin hydrochloride, the active ingredient in Glucophage 500mg, works through three complementary mechanisms. It reduces hepatic glucose production by inhibiting gluconeogenesis and glycogenolysis in the liver. It slows intestinal glucose absorption, reducing postprandial blood sugar spikes. It also improves peripheral insulin sensitivity, helping the body's own insulin work more effectively. Crucially, metformin does not stimulate insulin secretion directly, which means it carries a low risk of hypoglycaemia when used as monotherapy.


Important Information

Glucophage 500mg is a prescription-only medicine. Always consult your healthcare provider before starting, adjusting, or stopping this medication. Glucophage is contraindicated in patients with significant renal impairment (eGFR below 30 mL/min/1.73m2), hepatic impairment, acute metabolic acidosis, or dehydration. Renal function (eGFR) should be assessed before initiation and at least annually during treatment. A rare but serious risk - lactic acidosis - can occur if used inappropriately; seek immediate medical attention if you experience unusual muscle pain, difficulty breathing, or severe abdominal discomfort. Store at room temperature, away from moisture and direct sunlight.

Ingredients

Each white, circular, film-coated tablet is engraved with GL 500 and contains:

• Metformin hydrochloride 500mg (equivalent to 390mg metformin base)

• Excipients: Povidone, magnesium stearate, hypromellose (film coating)


Metformin belongs to the biguanide class of antihyperglycaemic agents. It is not chemically related to other oral antidiabetic drug classes and has a well-researched cardiovascular safety profile, with the landmark UK Prospective Diabetes Study (UKPDS) demonstrating a reduction in cardiovascular complications in overweight T2DM patients.



How to Use

Glucophage 500mg is taken with or immediately after meals. The usual starting dose for adults is 500mg two to three times daily, adjusted after 10 to 15 days based on blood glucose monitoring. The maximum recommended daily dose is 3g (six 500mg tablets). Starting at the lowest effective dose and increasing gradually helps minimise gastrointestinal side effects such as nausea, diarrhoea, or stomach discomfort, which typically subside as the body adjusts. This product is also used under medical supervision in polycystic ovary syndrome (PCOS) to address insulin resistance.
