Description
Janumet XR is a fixed-dose combination therapy containing Sitagliptin and Metformin Hydrochloride Extended-Release. This formulation provides physiological support for blood glucose management by utilizing two distinct mechanisms: Sitagliptin (a DPP-4 inhibitor) increases insulin synthesis and decreases glucagon secretion, while Metformin (a biguanide) reduces hepatic glucose production and improves insulin sensitivity.
Dosage & Administration
- 1 to 2 tablets per day, depending on the specific prescription regimen.
- Administer orally once daily.
- Timing: Must be taken with a meal, preferably in the evening, to reduce gastrointestinal side effects and maintain extended-release efficacy.
- Swallow whole: Do not split, crush, or chew. The extended-release matrix relies on an intact tablet structure for controlled delivery over 24 hours.
- Hydration: Administer with a full glass of water.
Safety & Storage
- Black Box Warning: Risk of Lactic Acidosis—a rare but serious metabolic complication associated with Metformin accumulation. Discontinue use before any radiologic study involving iodinated contrast.
- Adverse Reactions: May include diarrhea, nausea, and upper respiratory tract infection. Monitor for symptoms of pancreatitis or severe joint pain.
- Storage: Store in a dry environment at controlled room temperature (20°C to 25°C). Keep the container tightly closed to protect from moisture.
